چکیده

We wish to express our thanks to Dr Melvin Cohn for much valuable advice during the preliminary work on methods of purification, and one of us (M.R.P.) is greatly indebted to Dr Jacques Monod of the Institut Pasteur, Paris, for the hospitality of his laboratory in which much of the earlier work on isolation ofpenicillinase was carried out. We are also grateful to Dr T. S. Work for help in the chromatographic estimation of amino acids. The photographs were taken by Mr M. R. Young. The Perkin-Elmer Tiselius apparatus used for electrophoresis analysis was purchased by meauIs of a grant from Eli Lilly and Co.

The cell-bound penicillinase of Bacillus cereus.

Only 30-50 yo of the cell-bound penicillinase of Bacillus cereus NRRL 569 is neutralized by antiserum prepared against the exo-penicillinase. The unneutralizable fraction is not decreased by cell disintegration which liberates a proportion of the cell-bound enzyme into solution. Absence of neutralization cannot therefore be explained by the existence of a mechanical barrier which prevents acces...

New Assay for Penicillinase and Some Results on Penicillinase Induction.

Imsande, John (Western Reserve University, Cleveland, Ohio). New assay for penicillinase and some results on penicillinase induction. J. Bacteriol. 89:1322-1327. 1965.-A rapid, sensitive, and reliable assay of penicillinase activity is described. Studies conducted on the induction of penicillinase, with use of the new assay, show that actinomycin D inhibits growth and the induced synthesis of p...

Induction of penicillinase with inorganic phosphate.

Phosphate stimulates penicillinase formation in Bacillus cereus 569. The rate of penicillinase synthesis in the presence of 0.3 m phosphate, pH 7.0, is approximately 10-fold greater than that for uninduced cells, while the rate of synthesis in the presence of 0.3 m phosphate and 1 unit/ml of penicillin is approximately fourfold greater than in the presence of penicillin alone. When phosphate-in...

Penicillin and cell wall synthesis: a study of Bacillus cereus by electron microscopy.

The changes in wall structure of a penicillinase micro-constitutive strain of Bacillus cereus (569/H/24), on exposure to penicillin, and after its removal by addition of penicillinase, have suggested the following model for the growth of the walls of these cylindrical cells. Longitudinal extension is by addition of material to a large and continuously increasing number of growing points uniform...

Purification and Properties of the Exopenicillinase from Staphylococcus Aureus.

Resistance of Staphylococcus aureu8 to benzylpenicillin in clinical practice is usually caused by the production by the bacteria of an inducible penicillinase (EC 3.5.2.6) (Barber, 1962). The penicillinase of induced exponentially growing cultures of S. aureu8 is partitioned between the cells and the culture medium, the exact distribution varying from strain to strain and depending on the growt...
